ABSTRACT

OBJECTIVE Bergapten (BG), is a furanocoumarin derived from herbal and citrus extracts can act as antioxidant and selective anticancer agents. The current study aimed to investigate whether bergapten would attenuate immunosenescence and to exploreits immunomodulatory effects on immune responses in D-galactose-induced aging BALB/c mice.METHODS Firstly, mice were given D-galactose(180 mg·kg-1)subcutaneous injections for 30 d.To evaluate the establishment of the aging-related effect in mice, serum samples of BALB/c mice were collected from tail vein. Aging BALB/c mice were freely divided into three groups: negative control group received 1% Tween 80 solution only, named D-gal group. Positive groups were received BG administration at the dose of 20 and 100 mg·kg-1, named D-gal+BG(20)group and D-gal+BG(100)group,respectively.Effects of bergapten on T lympho-cyte proliferation and flow cytometry were assessed by using the splenic cell suspension. Enzyme linked immunospot kits were used to quantitatively determine interferon-γ(IFN-γ)and interleukin-4(IL-4) levels of the isolated serum. Immunophenotype was determined by using mixture of antibodies includ-ing anti-CD3,anti-CD4,and anti-CD8.RESULTS Bergapten(20 mg·kg-1)therapy can modulate immu-nity against viral epidemics and attenuate aging-induced immune deficiency(P<0.01),which was correlat-ed with the decline in the activation of the Th and Tc responses in D-galactose induced aging BALB/c mice.According to the in vivo results, bergapten exposure up-regulated the secretion of IFN-γ and IL-4 in T-helper 1(Th1)and T helper 2(Th2)cells(P<0.05,P<0.01).Additionally,BG(20 mg·kg-1)restored antigen-specific CD4+and CD8+T cells in aging models (P<0.05, P<0.01), which may help to curing chronic infections. CONCLUSION The beneficial effect of bergapten in D-galactose induced aging BALB/c mice may be due to the Th and Tc responses activation.

ABSTRACT

BACKGROUNDDespite extensive research, the mechanism of immature dendritic cells (DCs) induced immune hyporesponsiveness remains incompletely understood.

METHODSRecipient DCs from C3H mouse bone marrow cells were incubated with donor antigen from splenic lymphocytes of C57BL/6 mouse; these DCs were transfected with CD80/86 specific siRNA using lentiviral vectors. Flow cytometry was used to evaluate expression of CD80/86 on the antigen-pulsed recipient DCs. Immune regulatory activity was examined by mixed lymphocyte reaction, in which irradiated DCs were cultured with C3H spleen T cells. After the reaction, interleukin (IL)-2, IL-4, IL-10, and interferon (INF)-γ levels of mixed lymphocyte reaction culture supernatant were measured by enzyme-linked immunosorbent assay. The apoptotic T lymphocytes were identified by Annexin V and CD3 staining.

RESULTSThere was a significant inhibition of CD80/86 expression in DCs transfected with CD80/86 lentiviral vectors compared with the control groups (P < 0.05), indicating the specificity of RNA interference. Enzyme-linked immunosorbent assay results showed a significant reduction of INF-γ, IL-2 and IL-10 in the CD80/86 lentivirus transfected group compared to the control groups (P < 0.05). There was no significant difference in IL-4 levels between the groups (P > 0.05). We also showed that CD80/86 low DCs loaded with alloantigen (1) stimulated low T cell proliferative responses via the indirect recognition pathway and (2) enhanced apoptotic activity (P < 0.05) in co-cultured T cells.

CONCLUSIONSLentiviral vector transfection can effectively and specifically knock down target genes in DCs. The CD80/86 low DCs may show tolerogenic activity via induction of T-cell apoptosis, thereby modulating the activity of recipient-derived DCs. The use of this approach may potentially be clinically applicable.

ABSTRACT

Objective To investigate the preliminary clinical application of anterior pedicle screws in treatment of the lower cervical spine injuries. Methods The study involved 22 patients with lower cervical spine injuries treated with anterior pedicle screw technique from January 2009 to December 2011.X-ray and CT images of the patients were taken postoperatively to evaluate whether the position, orientation and length of the screws were appropriate and whether there involved pedicle cortex perforation, transverse foramen or spinal canal invasion.MRI was also performed to ascertain the situation of decompression, the formation of epidural hematoma and the degeneration of cervical spinal cord. The improvement of JOA score was compared statistically. Results All the patients were followed up for 3-36 months ( mean,15.5 months).A total of 44 anterior lower cervical screws were implanted and all screws were inserted smoothly. All the patients had bone healing after average 4.5 months, which showed no loosening, prolapse or breakage of the screws or no loosening of the plate. One patient showed hoarseness postoperatively, which was probably due to the long-term distraction of recurrent laryngeal nerve. The symptom disappeared at around three weeks after neurotrophic support. Two patients complained of dysphagia postoperatively, and were informed of keeping a soft diet. The symptom was remarkably alleviated after three months or so.The X-ray films indicated satisfactory position of the implanted screws. Before discharge, the transaxial CT images showed that two screws perforated the medial pedicle cortex ( 1 ° ) and that two screws perforated the lateral cortex (1 °) and affected the medial margin of the transverse foramen. Before discharge, the MRI indicated obvious alleviation of the anterior spinal cord compression in all patients and a small amount of anterior epidural hematoma in four patients, with no obvious spinal cord compression. At 12 months after operation, the MRI showed significant improvement of spinal cord degeneration in three patients.JOA score was increased from preoperative (8.5 ± 0.7) to ( 14.5 ± 0.8 ) before discharge (P <0.01 ).JOA score was (15.7 ±0.7) at six months postoperatively, which was significantly improved as compared with that before discharge. Conclusions Anterior pedicle screw fixation is a reliable and safe method for reconstruction of the anterior lower cervical injuries.Surgical indications should be strictly controlled in its clinical application.

ABSTRACT

OBJECTIVETo compare three techniques of the transarticular screw placement in the lower cervical spine.

METHODSThe techniques of transarticular screw placements used by Takayasu (group A), Dalcanto (group B) and Klekamp (group C) were applied in twenty-four cervical specimens, among which 14 specimens were male and 10 specimens were female, ranging in age from 55 to 80 years, with an average of 68 years. The entry point of Takayasu technique was between the superior and median third of the vertical medial line of the lateral mass with 60 to 80 degrees caudal tilt and 0 degrees lateral tilt. Dalcanto technique started 2 mm caudal to the midpoint of lateral mass with 40 degrees caudal tilt and 20 degrees lateral tilt. Klekamp technique inserted the screws with the starting point being 1 mm medial and 1 mm inferior to the midpoint of the lateral mass with 40 degrees caudal tilt and 20 degrees lateral tilt. The splits of facets, the encroachment of the cervical nerve roots and vertebral arteries, and no-involving facets screws were observed and analyzed.

RESULTSOne hundred and ninety-two transarticular screws were implanted on both sides in twenty-four cervical cadavers, sixty-four for each group. There were twenty-five splits of inferior facets in group B, 2 splits in group C and none in group A. No superior facets were found to be broken in all cases. The difference of splits between group B and the other two groups was significant (Chi2AB = 31.07, P < .001; Chi2BC = 24.83, P < 0.01), while there was no statistical difference between group A and C (Chi2AC = 2.03, P > 0.05). Thirty-six screws encroached the vertebral arteries in group A, none in group B and C. There was significant statistical difference between group A and the other two groups (Chi2AB = 50.09, P < 0.01; Chi2AC = 50.09, P < 0.01). Forty anterior branches of lower cervical nerve roots were involved in group A, five in group B and three in group C. There were statistical differences between group A and the other two groups (Chi2AB = 41.98, P < 0.01; Chi2AC = 47-94, P < 0.01), and there was no statistical difference between group B and C. Sixteen posterior branches of lower cervical nerve roots were encroached in group A, 18 in group B and 14 in group C. There was statistical difference among the three groups (Chi2AB = 0.16, P > 0.05; Chi2AC = 0.17, 0.05; Chi2 = BC = 0.67, P >0.0 ). Although all screws went through facets in the study except for 5 in group A, there were no statistical differences between each groups Chi2AB k =3.33, P >0.05; X2A C =3.33, P> 0.05).

CONCLUSIONThere are high risk of injury of anterior branch of cervical nerve root and vertebral artery if the screws are too long and the Takayasu's technique is used. However, the rate of facet split is high if the Dalcanto's technique is applied. Klekamp's technique is value to be applied.

ABSTRACT

OBJECTIVETo study the conditions and parameters of purifying total flavonoids from seed residue of H. rhamnoides.

METHODAbsorption capacity of three resins for total flavonoids was compared. With the yield and purity as indexes, the process of absorbing and purifying total flavonoids from seed residue with D101 macroporous resin absorbent was selected by orthogonal design.

RESULTThe D101 resin was the best of the three resins and its absorption capacity was determined to be 63.00 mg x g(-1) wet resin. The optimum process condition was 30% ethanol as eluting solvent, wet resin vs crude herbs: 2:1, diameter vs height: 1:10, eluting solvent vs crude herbs: 10:1, absorbing time for 3 h. The yield of total flavonoids from seeds residue of H. rhamnoides by this process was 2.39%, and the purity reached 64.81%.

CONCLUSIONThe process is simple and convenient and the regeneration of resin is easy. So this method of purification is advisable.
